Öğe Anti-aging and Antioxidant Activities of Ethanol and Three-phase Partitioned Extracts of Inula viscosa(Amg Transcend Assoc, 2023) Uckaya, FatihInula viscosa is a plant commonly used to treat inflammation, germicide, expectorant, cough reducer, diuretic, anemia, and cancer. In addition to its medicinal effect, this plant has been preferred because it is thought to be a free radical scavenger and anti-aging effect due to its phenolic compounds. The aim of the study was to determine whether the plant has antioxidant and anti-aging potential. The plant was extracted with both the traditional solvent extraction method with ethanol and the novel three-phase partitioned (TPP) system, and the two methods were compared. The methods of DPPH free radical scavenging, ABTS cation radical scavenging, superoxide anion radical scavenging, and reduction of copper and iron ions were used to determine the antioxidant effects of the obtained extracts. In addition, elastase enzyme inhibition was studied to investigate the anti-aging potential. TPP method (23.9% yield) was performed at a higher yield than the solvent extraction (8.99% yield). The ethanolic extract and TPP extract of plant inhibited the DPPH free radical with IC50=0.27 mu g/mL and IC50=0.026 mu g/mL, and ABTS cation radical with IC50=0.28 mu g/mL and IC50=0.13 mu g/mL, and superoxide anion radical with IC50=3.53 mu g/mL and IC50=2.43 mu g/mL, respectively. According to the elastase inhibition results, IC50=3.11 mu g/mL for ethanol extract and IC50=3.05 mu g/mL for TPP extract were obtained. Both extracts of I. viscosa showed antioxidant and anti-aging effects at levels close to the standards. In addition, according to the results, TPP extract was found to be more effective than ethanol extract.Öğe In Vitro Anti-Aging Activitiy of Arum Dioscoridis with In Silico Prediction of Its Active Compounds(Chem Soc Pakistan, 2024) Uckaya, FatihArum dioscoridis is a plant that has been used by people for centuries for shortness of breath, asthma, bronchitis, expectorant, intestinal laziness, digestive system, malaria and especially hemorrhoid diseases. The aims of the study are to determine the efficient extraction method by extracting the plant with different methods, to investigate the antioxidant and anti-aging effects of the extracts, and to perform in silico prediction analyzes of the active molecules of the plant. The plant was extracted using ethanol by maceration, ultrasonic wave extraction, and soxhlet extraction. The antioxidant and anti-aging potential of the obtained extracts were investigated. To determine antioxidant activity, DPPH free radical scavenging, cupric ion reduction and iron ion reduction methods were applied. Anti-aging activity was determined by inhibiting the enzyme elastase. Additionally, the active components of the ethanol extract of the plant were determined using PASSonline, LAZAR, ADMETlab 2.0 and CLC-pred online prediction programs. The yields obtained from the maceration, ultrasonic wave extraction and soxhlet extraction methods are 8.99 g/100g, 19.94 g/100g and 13.2 g/100g, respectively. The IC50 values from the DPPH method obtained from the maceration ethanol extract, ultrasonic wave ethanol extract, and soxhlet ethanol extract were 3.943 mu g/mL, 2.797 mu g/mL, and 2.273 mu g/mL, respectively. The IC50 values of the extracts obtained from the elastase inhibition method were 3.112 mu g/mL, 3.357 mu g/mL and 2.964 mu g/mL, in the same order. Additionally, the extracts showed inhibition of cupric ions at 1.8 nm, 0.81 nm and 1.79 nm, in the same order. In silico biological activity was determined with the PASSonline program, toxicity with LAZAR and ADMETlab programs, and the effects of active molecules against some cell lines were determined with the CLC-Pred program. As a result, it was determined that the plant has biological activities to protect the skin against aging in silico and in vitro.Öğe Prangos ferulacea (L.) Lindl. Bitkisinin Toplam Fenolik ve Toplam Flavonoit Madde Miktarı ve Antioksidan Aktivitesinin Belirlenmesi(2023) Hancer, Cagla; Yavuz, Oğuzhan; Uckaya, FatihPrangos ferulacea (L). Lindl. Erzurum ili ve çevrelerinde halk arasında tıbbi ve gıda amaçlı olarak kullanılan bir bitkidir. Özellikle şehrin kuzey taraflarında her yılın Mayıs ayında toplanan P. ferulacea (Yöresel adı Çaşır, Çakşır) yaprakları salamura yapılarak tüm yıl boyunca gıda olarak tüketilmektedir. Bu çalışmada Mayıs ayında araziden toplanan bitkinin yaprakları kurutulup 6 farklı çözücü ile (su, metanol, %70 metanol, hekzan, kloroform, aseton) ekstresi, uçucu yağı ve yağ altı suyu elde edilmiştir. Bitkinin salamurasından ise 4 farklı çözücü (aseton, metanol, hekzan, klorofom) ile ekstresi elde edilmiştir. Tüm ekstrelerin antioksidan aktivite ve toplam fenolik-flavonoid madde içeriğine bakılmıştır. Ayrıca deney gruplarından elde edilen verilerin temel bileşen analizi (PCA) ve CLUSTVIS yardımı ile ısı haritası kümelemesi yapılmıştır. Sonuç olarak P. ferulacea yapraklarından hazırlanan ekstrelerin; DPPH yönteminde metanol, su ve %70 metanol kuru bitki ekstrelerinin; ABTS yönteminde %100 metanol, %70 metanol ve su kuru bitki ekstrelerinin standart antioksidanlar kadar etkili oldukları görülmüştür. CUPRAC yönteminde ise kuru bitki su ekstresi 100 ?g/mL konsantrasyonunda 1,86 absorbans değeri ile en yüksek bakır iyonu indirgeme gücüne sahip ekstre olmuştur. Kuru bitki metanol ve salamura hekzan ekstreleri 100 ?g/mL konsantrasyonunda 1 absorbans değerinden daha yüksek etki göstermiştir. Bitkiden elde edilen uçucu yağ ve yağ altı suyu her 3 yöntemde de en düşük antioksidan aktiviteyi göstermiştir. Toplam fenolik madde içeriği incelendiğinde su, aseton ve metanol kuru bitki ekstrelerinin; toplam flavonoid madde içeriği incelendiğinde ise klorofom ve su kuru bitki ekstreleri ile metanol salamura ekstresinin en yüksek madde içeriğine sahip oldukları görülmüştür. Özellikle yaprakları gıda olarak kullanıldığı için antioksidan aktivite değerlendirmesi açısından su ekstresinin her 3 yöntemde de iyi sonuçlar vermesi, ayrıca toplam fenolik- flavonoid madde içeriğinin yüksek değerleri P. ferulacea bitkisinin gıda olarak kullanım şeklini desteklemektedir.
