Öğe Comparison of Craniosacral Therapy and Myofascial Relaxation Techniques in People with Migraine Headache: A Randomized Controlled Study(Elsevier Inc, 2024) Araci, Ayca; Ozsimsek, Ahmet; Yulug, Burak; Karacay, ErtanObjective: The primary objective of this study was to investigate the impact of myofascial release and craniosacral therapy on the quality of life, pain levels, and range of motion (ROM) in patients with chronic migraine headaches. The secondary aim of this study was to develop a migraine treatment protocol using current craniosacral techniques. Methods: Patients with chronic migraine in the Neurology Department of ALKU Hospital were randomly allocated to 3 therapy groups: (1) Craniosacral Treatment Craniosacral Techniques (CST) + Medical Treatment (MT) (CST group) (n = 24), (2) myofascial treatment (MFT) (n = 24) + MT (MFT group), and (3) MT (control group) only (n = 26). Visual Analog Scale (VAS) for pain, FONSECA for temporomandibular disorder symptom intensity, 24 Hours Quality of Life Questionnaire Scales for quality of life, and Migraine Disability Assessment Score for impairment, Goniometer for Cervical ROM were used for the disability level. Follow-up scores were collected 4 times: at pretreatment (T0), immediately post-treatment (T1), 1 month (T2), and 3 months after treatment ended (T3). Results: Changes were found in T0 to T1 treatment results, VAS, and ROM angles between the groups. In intragroup evaluations, 24 Hours Quality of Life Questionnaire changes were observed only in the CST group at T0 to T1 to T2 periods (P = .011) while Migraine Disability Assessment Score scores were significantly changed in all groups. Significant changes were also observed in both VAS scores and FONSECA scores of the CST and MFT groups whereas VAS scores decreased significantly, especially in the T0 to T1 to T2 to T3 periods (P < .05). In the evaluation of FONSECA scores both within and between groups, it was observed that the most significant decrease was in the T2 period and there was a difference between the groups (P = .015). Conclusion: For the participants in this study, CST and MFT techniques reduced migraine headache, temporomandibular disorder level, drug consumption, and functional disability levels, and increased cervical region ROM. These results suggest that CST techniques could be considered in migraine treatment as one of the clinical practical applications within the framework of a certain protocol. (J Chiropr Med 2024;23;114-126)Öğe Ginkgo Biloba Özütünün in Vitro Alzheimer Hastalığı Modelinde TRPV1 Kanalları Aracılığıyla Koruyucu ve Terapötik Rolü(2023) Özşimşek, Ahmet; Övey, İshak Suat; Karacay, Ertan; Yuluğ, BurakAmaç: Ginkgo biloba’nın (GB) nöroblastom hücrelerinde mitokondriye bağımlı TRPV1 iyon kanalları üzerindeki etkisi Alzheimer hastalığı (AH) modeli oluşturularak araştırıldı. Yöntem: Bir AH modeli oluşturmak için SH-SY5Y hücrelerine okadaik asit uygulandı. Hücresel farklılaşmadan sonra yedi ana grupla organize edilen çalışma, GB’nin nöroblastom hücrelerinde AH’de kalsiyuma bağlı TRPV1 kanallarına etkisinin incelendiği in vitro çalışmadır. Bulgular: Kontrol grubu ile karşılaştırıldığında GB+AH, AH ve AH+GB gruplarında daha yüksek Ca 2+ konsantrasyonu saptandı (p<0,001). Ca 2+ düzeyi GB+AH ve AH+GB gruplarında AH grubuna göre daha düşüktü (p<0,001). Ayrıca GB+AH’de sitozolik Ca 2+ konsantrasyonu AH+GB grubuna göre daha düşüktü (p<0,05), apoptoz ve hücre içi reaktif oksijen türleri (ROS) değerleri GB+AH, AH ve AH+GB’de control grubuna göre daha yüksekti (p<0,001). Apoptoz ve hücre içi ROS değerleri AH grubunda GB+AH ve AH+GB grubuna göre daha yüksekti (p<0,001) ve AH+GB grubunda apoptoz düzeyi GB+AH grubuna göre daha yüksekti (p<0,001) ve mitokondriyal depolarizasyon, kaspaz 3 ve kaspaz 9 düzeyleri GB+AH, AH ve AH+GB gruplarında kontrol grubuna göre daha yüksekti (p<0,001). Ayrıca GB+AH+kapsazepin grubu, AH+kapsazepin grubu ve AH+GB+kapsazepin ile karşılaştırıldığında GB+AH grubu, AH grubu ve AH+GB gruplarında değerler daha düşüktü (p<0,001). Sonuç: Bu sonuçlar bize GB’nin TRPV1 kanalı üzerinden Alzheimer hastalığında tedavi edici etkisinin yanında koruyucu etkisinin olduğunu göstermektedir.Öğe Protective and Therapeutic Role of Ginkgo Biloba Extract Through TRPV1 Channels in an in Vitro Alzheimer's Disease Model(Turkish Neuropsychiatry Assoc-Turk Noropsikiyatri Dernegi, 2023) Ozsimsek, Ahmet; Ovey, Ishak Suat; Karacay, Ertan; Yulug, BurakIntroduction: The effect of Ginkgo biloba (GB) on mitochondria dependent TRPV1 ion channels in neuroblastoma cells was investigated by creating an Alzheimer's disease (AD) model.Methods: Okadaic acid was applied on SH-SY5Y cells to create an AD model. After cellular differentiation, the study was organized with the seven main groups, examining the effect of GB on calcium depended TRPV1 channels in neuroblastoma cells AD, has been established in vitro.Results: The higher Ca2+ concentration was detected in the GB+AD, AD and AD+GB groups when compared with the control (p<0.001). The Ca2+ level was lower in GB+AD and AD+GB groups than in the AD group (p<0.001). Also, cytosolic Ca2+ concentration was lower in the GB+AD than in the AD+GB group (p<0.05), the apoptosis and intracellular reactive oxygen species (ROS) values were higher in the GB+AD, AD and AD+GB groups than in the control (p<0.001). The apoptosis and intracellular ROS values were higher in AD group than in the GB+AD and AD+GB group (p<0.001) and the apoptosis level was higher in AD+GB group than GB+AD group (p<0.001) and the mitochondrial depolarization, caspase 3 and caspase 9 levels were higher in the GB+AD, AD and AD+GB groups when compared to the control group (p<0.001). Also, the values were lower in the GB+AD group, AD group and AD+GB groups when compared with the GB+AD+capsazepine group, AD+capsazepine group and AD+GB+capsazepine respectively (p<0.001).Conclusion: These results show us that GB has a protective effect besides its therapeutic effect in Alzheimer's disease via TRPV1 channel.
